A theoretical compound thats estimated to be in the range of 4,000 to 26,600x as potent as diazepam, or 400-1,300x as potent as alprazolam. 0.375-2.5ug could be equivelent to 10mg of diazepam. I call ot diflunitizolam. What makes it so potent is mainly the 2,6-difluorophenyl ring. A second fluorine atom bonded into the 6 position of the phenyl ring causes it to twist into the perfect position for the subpocket of the bzd site. My estimate comes from known benzo analogs, like flunitrazolam in which a threshold dose is 50-100ug. The thiophene substitution is known to increase potency by roughly 2-4x and the dliflurophenyl substitution is also known to boost the potency by 5-10x on top of everything. Its binding affinity goes from being measured in nanomolars into picomolars. This would be the limit to how potent a benzodiazepine could be. Its half life is estimated to be 6-12 hours.
