For pretty much my entire experience using substances and talking to people about substances, i have heard the common “only water soluble drugs can be absorbed well in nasal mucosa”. I bet most of you have heard this as well. The thing that interests me the most is how common this belief is and how utterly wrong it is. Lipophilic/hydrophobic molecules are far more bioavailable insufflated than lipophobic/hydrophilic molecules. The only explanation i have seen is when people are discussing why xanax/alprazolam isnt able to be snorted. They claim because xanax is hydrophobic and has horrible bioavailability in nasal mucosa that “drugs need to be hydrophilic to be absorbed well.” But that’s literally the only “explanation” ive seen even though it’s not a very good one. While having slight water solubility is helpful when snorting a drug, due to nasal secretions being mostly water, its far more important for a drug to be lipophilic in order for it to cross the lipid bilayer/ blood brain barrier, in order for it to be effective.

When you look at drugs that are highly water soluble, you can easily determine that fat solubility is far more important. Morphine for example: yes morphine is absorbed through nasal mucosa, but it’s bioavailability is extremely poor (10%-17%).

Its far more complex than “a drug needs to be lipophilic” or “a drug needs to be hydrophilic) in order to be absorbed well, but the former is more important simply because of fat soluble molecules crossing the bbb effectively. For some drugs, being fat soluble in freebase form, and being a little more hydrophilic when in salt form can be helpful, but for the most part, freebase fat soluble compounds that have a small molecular structure are going to be most effective when snorted.
